The habenulo-interpeduncular (Hb-IPN) conduction system is a highly conserved neural pathway involved in diverse physiological and pathological functions, such as stress, fear and nicotine withdrawal symptoms. The Hb-IPN tract is a major cholinergic system in the mammalian brain. In zebrafish, the Hb-IPN system has also emerged as a powerful model to study brain laterality, as the left-right habenulae exhibit striking differences in their morphology, molecular properties and connectivity. Through the discovery of a second cholineacetyltransferase locus, we identified distinct cholinergic neuronal populations in the larval zebrafish brain. Interestingly in the habenulae, cholinergic neurons are present mainly in the right nucleus, suggesting that there is functional specialization in this region of the larval brain. Left (non-cholinergic) and right (cholinergic) habenular neurons innervate different domains of the IPN and likely activate different downstream targets. Adult fish exposed to nicotine exhibit increased fos expression in IPN neurons that express a specific nicotinic acetylcholine receptor subunit, suggesting that these neurons respond to nicotine. Overall, this work provides strong evidence for the conservation of cholinergic systems in the vertebrate brain and provides a framework to start dissecting novel neural pathways that become preferentially altered by nicotine addiction.
