Searching for a novel obesity biomarker :
Proteomic analysis of hepatic steatosis from high-fat diet induced obese mice
Overweight and obesity are defined as abnormal or excessive fat accumulation. They are major risk factors for a number of chronic diseases such as diabetes, cardiovascular diseases, hypertension, and certain cancers. The molecular mechanisms involved in obesity are still unclear. It is widely accepted that disruption in the homeostasis between food intake and energy expenditure, and the subsequent deposition of excess fatty acids into fat cells in the form of triglycerides, leads to the development of obesity. To identify proteins potentially involved in obesity processes, we performed a proteomic analysis of obesity-induced mouse liver. Male C57BL/6J mice fed a high-fat diet (HFD) for 24 weeks developed hepatic steatosis characterized by significantly increased free fatty acid (FFA) and triglyceride levels. 2D-based proteomic analysis revealed that, compared the normal diet (ND), HFD changed the expression of many proteins. The most pronounced difference was intermediate microfilament (IF) cytoskeleton proteins. In particular, vimentin as well as cytokeratins (CK-8 and CK-18) was significantly upregulated in obese animals. This suggests a significant alteration in IF organization, which may be in part responsible for the development of obesity and steatosis. The expression of some selected proteins was validated by Western blot, microarray analysis and hepatocyte morphology by immunohistochemistry. The results demonstrated that overexpression of IFs in liver is related to obesity and the level may be a useful marker for predicting obesity and liver disease.
